Abstract

Using a simple High-Performance Liquid Chromatographic (HPLC) procedure, the pharmacokinetic parameters of oral diazoxide 100 mg capsule (Razak company) and proglycem® (Shering Company) in male volunteers were evaluated. Plasma samples were injected directly into the HPLC only after deproteination by perchloric acid. The chromatographic separation was accomplished with a very simple mobile phase consisting 40 methanol in water at a flow rate of 1.5 ml/min using spherisorb C18 column and ultraviolet detector at a wavelength of 270 nm. This method was sensitive with a quantification limit of approximately 0.1μg/ml. The linearity was assessed in the range of 2-200 ng with a correlation coefficient of greater than 0.986. The concentrations versus time curve of both compounds were superimposed except at the peak concentration. These results indicate that the Razak 100 mg Diazoxide capsule is bioequivalant with proglycem®. Results of this study showed, use of a simple plasma deproteination step instead of extraction was sensitive, easy, rapid and precise for determination of diazoxide.
